I have upgraded to WordPress 2.5.1, in a few easy steps based on the Codex article:

  1. Backup database (using phpMyAdmin, in my case)
  2. Backup wp-contents folder (where I have my themes and plugins)
  3. Deactivate all the plugins
  4. Delete wp-includes and wp-admin folder
  5. Upload new WordPress files and overwrite the old ones
  6. If you lost your wp-contents folder, you can always re-upload it using the backup you had.
  7. Go to http://yoururl/wp-admin/upgrade.php to complete the upgrade
